He would most likely have been detained and used as an experimental subject after hospital tests revealed blood anomalies or cellular mutation.

That was the 1960s, if he went to the hospital his bill would have been about $3.00.  I saw my mothers bill for when she gave birth to my brother and it had at least 2 days in the hospital and that was $45.00.    Remember, Peter used to take pictures to pay for Aunt May's long hospital stays.  One spider bite would be nothing.

What If 7 had a story like this, where some of the supporting cast got the powers, but with negative outcomes, apart from Parker continuing as the only Spider-Man in the end.

But, the arachnid powers have always found their way in isolation to others; Jessica Drew, Spider-Gwen, Miles Morales, etc.
